Takahashi, I. (McMaster University, Hamilton, Ontario, Canada). Transduction of sporogenesis in Bacillus subtilis. J. Bacteriol. 89:294-298. 1965.-A number of asporogenous mutants of Bacillus subtilis were isolated and genetic analyses with these mutants were carried out by means of transduction mediated by bacteriophage PBS 1. The ability to form spores could be restored by transduction in all mutants examined, with the exception of Sp(-)H12-3, although they were not transformable under my experimental conditions. The results of reciprocal transduction suggest that genetic loci at which a mutation can affect sporulation are numerous. A spore marker (sp 1) was transduced jointly with prototrophy and some antibiotic resistance markers (streptomycin, erythromycin, and neomycin). Other spore markers (sp N2-2, sp 170-2, and sp H12-4) were also linked to auxotrophic markers (ser, tyr, and phe) at a fairly high frequency. This linkage relationship indicates that genes controlling the formation of spores are located along the chromosome far apart from each other. The result also suggests that blocks in a variety of biochemical processes may result in the expression of the asporogenous phenotype.

Induction of Bacillus subtilis sporulation by nucleosides: inosine appears to be sporogen.

Inosine completely reversed the selective inhibition of sporulation in Bacillus subtilis 168 caused bym-aminobenzeneboronic acid; guanosine and adenosine, but not xanthosine, partially reversed inhibition, whereas pyrimidine nucleosides were slightly effective. In addition, 0.005 to 0.025 mM inosine caused a four- to fivefold stimulation of sporulation of B. subtilis grown in minimal salts medi...

The Oxidative Metabolism of Bacillus

The genus BaciUus is one of the few remaining important groups of bacteria for which a route of terminal oxidation has not been shown. In studies (Gary et al., 1954) on the effect of nutrition on metabolic patterns in Bacilus subtilis, it was suggested that cells grown in a glucose mineral salts medium probably possefs the enzymatic mechanisms of the tricarboxylic acid cycle. Hardwick and Foste...

Isolation and sequence of ctaA, a gene required for cytochrome aa3 biosynthesis and sporulation in Bacillus subtilis.

Cytochrome aa3 is one of two terminal oxidase complexes in the Bacillus subtilis electron transport chain. A novel genetic strategy was devised which permitted the isolation of B. subtilis mutants lacking cytochrome aa3 by selection for streptomycin-resistant clones which failed to oxidize the artificial electron donor N,N,N',N'-tetramethyl-p-phenylenediamine. Two mutations were studied intensi...

Bacillus subtilis bacteriophage SPbeta: localization of the prophage attachment site, and specialized transduction.

The attachment site for the prophage of SPbeta lies between ilvA and kauA on the chromosome of Bacillus subtilis strain 168. Specialized transduction of citK and kauA can be carried out by certain lysates of SPbeta.

A Novel Cell Type Enables B. subtilis to Escape from Unsuccessful Sporulation in Minimal Medium

Sporulation is the most enduring survival strategy developed by several bacterial species. However, spore development of the model organism Bacillus subtilis has mainly been studied by means of media or conditions optimized for the induction of sporogenesis. Here, I show that during prolonged growth during stationary phase in minimal medium, B. subtilis undergoes an asymmetric cell division tha...
